چکیده

Fibroblasts derived from the involved skin of scleroderma patients frequently display a phenotype of supernormal collagen expression when cultured. Fibroblasts displaying this phenotype derived from seven patients were treated with r~laxin (1-100 ngjml) and interferon-y (1-100 U jml), individually and in combination, to assess the relative abilities of these cytokines to down-modulate collagen synthesis and S~cretion. Scleroderma fibroblasts displayed varying sensitivities to both relaxin and interferon-yo Relaxin (100 ng/ml) decreased expression of collagen by six of seven lines tested from 8 to 59% compared to untreated cultures. Interferon-y (100 U jml) depressed collagen secretion by all seven lines in

In vitro Co-Culture of Human Skin Keratinocytes and Fibroblasts on a Biocompatible and Biodegradable Scaffold

Background: Extensive full-thickness burns require replacement of both epidermis and dermis. In designing skin replacements, the goal has been to re-create this model and make a product which has both essential components. Methods: In the present study, we developed procedures for establishing confluent, stratified layers of cultured human keratinocytes on the surface of modified collagen-chito...

The Effects of Low Level Laser Therapy on the Expression of Collagen Type I Gene and Proliferation of Human Gingival Fibroblasts (Hgf3-Pi 53): in vitro Study

Objective(s): Recent investigations show that both proliferation and secretion of macromolecules by cells can be regulated by low level laser therapy (LLLT). The aim of this study was to determine whether LLLT could induce a bio-stimulatory effects on human gingival fibroblasts (HGF3-PI 53). Therefore, the effect of laser irradiation on human gingival cell proliferation and collage...

Collagen synthesis by human fibroblasts

We have examined the combined effects of transforming growth factor-fl (TGF-,8), serum and y-interferon (y-IFN) on collagen synthesis by fibroblasts and compared the response of fibroblast subpopulations to TGF-,6. Human diploid fibroblasts were treated with TGF-/J alone and with serum or y-IFN. Cells were labelled with radioactive amino acids, and collagen production was measured as collagenas...
